摘要 采用高效液相色谱法测定植物油中3种生育酚(α-生育酚、β-生育酚、δ-生育酚)的含量.系统考察了流动相的种类、配比、柱温等因素对3种生育酚分离分析的影响.采用十八烷基硅烷键合硅胶为填充剂的色谱柱作为分析柱,体积分数100%的甲醇为流动相,在流速1 mL/min、柱温25 ℃、紫外检测波长292 nm的色谱条件下,α-生育酚、β-生育酚、δ-生育酚的分析时间在7 min内.α-生育酚、β-生育酚、δ-生育酚在质量浓度为5~120 mg/L范围内与峰面积呈良好线性关系(R2≥0.999 7).α-生育酚、β-生育酚、δ-生育酚的检出限(S/N=3)分别为0.2 mg/L、0.6 mg/L、0.3 mg/L,定量限(S/N=10)分别为0.8 mg/L、1.4 mg/L、1.0 mg/L.样品的加标回收率为96.6%~101.6%,RSD为0.9%~2.0%.该方法操作简单、快速、准确可靠,将其应用于植物油真实样品中3种生育酚含量的测定,获得了令人满意的结果.

Abstract: The contents of three kinds of tocopherols(α-tocopherol,β-tocopherol and δ-tocopherol)in vegetable oil were determined by HPLC.The influence of factors such as the type,ratio,and column temperature of the mobile phase on the separation and analysis of three types of tocopherols were investigated.A chromatographic column with octadecyl silane bonded silica gel as the filler was used as the analysis column,and methanol with a volume fraction of 100% was used as the mobile phase.With methanol as the mobile phase at the flow rate of 10 mL/min.The temperature of column is set at 25 .The detector is set at wavelength of 292 nm.The analysis time of α-tocopherol,β-tocopherol,and δ-tocopherol was within 7 minutes.Good linear relationships were obtained in the range of 5~120 mg/L and the correiation coefficient(R2)were all higher than 0999 6.The limits of detection(S/N=3)were 0.2 mg/L,0.6 mg/L and 0.3 mg/L,respectively,and the limits of quantification(S/N=10)were 0.8 mg/L,1.4 mg/L and 1.0 mg/L,respectively.The recoveries ranged from 96.6% to 101.6%,with the relative standard deviation(RSD)between 0.9% and 2.0%.The method is simple,rapid,accurate and reliable and has been applied to the determination of three kinds of tocopherols in real vegetable oil samples and obtained satisfactory results.
