Abstract The contents of three kinds of tocopherols(α-tocopherol,β-tocopherol and δ-tocopherol)in vegetable oil were determined by HPLC.The influence of factors such as the type,ratio,and column temperature of the mobile phase on the separation and analysis of three types of tocopherols were investigated.A chromatographic column with octadecyl silane bonded silica gel as the filler was used as the analysis column,and methanol with a volume fraction of 100% was used as the mobile phase.With methanol as the mobile phase at the flow rate of 10 mL/min.The temperature of column is set at 25 ℃.The detector is set at wavelength of 292 nm.The analysis time of α-tocopherol,β-tocopherol,and δ-tocopherol was within 7 minutes.Good linear relationships were obtained in the range of 5~120 mg/L and the correiation coefficient(R2)were all higher than 0999 6.The limits of detection(S/N=3)were 0.2 mg/L,0.6 mg/L and 0.3 mg/L,respectively,and the limits of quantification(S/N=10)were 0.8 mg/L,1.4 mg/L and 1.0 mg/L,respectively.The recoveries ranged from 96.6% to 101.6%,with the relative standard deviation(RSD)between 0.9% and 2.0%.The method is simple,rapid,accurate and reliable and has been applied to the determination of three kinds of tocopherols in real vegetable oil samples and obtained satisfactory results.
